چکیده

Group transfer reactions catalyzed by enzymes have assumed a position of intense interest in contemporary enzymology (1). Well known examples of radicals capable of being transferred in such reactions are methyl, glycosyl, aeetyl, phosphoryl, pept, idyl, nucleosidyl, and nucleotidyl, in addition to the transport of hydrogen as seen with the familiar nuoleot.ide coenzymes. In this paper we wish to report experimental evidence which establishes t,he phenomenon of an efficient enzyme-catalyzed transfer of the glucuronyl radical from a variety of donor substrates to a number of acceptor alcohols. This is accomplished by a soluble, stable, highly purified cell-free enzyme from liver which is in many respects indistinguishable from @-glucuronidase. These data supplement and extend the observations first reported in 1956 (2).

Catalysis of phosphoryl transfer by prostatic acid phosphatase.

As part of a study on enzymic catalysis of group transfer, this laboratory has previously reported on the enzymic transfer of the glucuronyl radical from a variety of donor substrates to a number of acceptor alcohols (1). The phenomenon was studied in detail and conditions for efficient group transfer were investigated. The present paper reports the use of a similar approach in studying the aci...

Relation between enzymic catalysis and energy coupling.

The principles that underlie enzyme catalysis also apply to energy coupling processes. A comparison is made between a kinase system that mediates the phosphorylation of glucose by ATP (hexokinase), as the prototype for enzymic catalysis, and the mitochondrial electron-transfer complexes, as the prototypes for energy coupling systems. Induced polarization of chemical bonds and charge separation ...

Reduction of intrinsic kinetic and thermodynamic barriers for enzyme-catalysed proton transfers from carbon acid substrates.

Many enzymes catalyse the heterolytic abstraction of the alpha-proton from a carbon acid substrate. Gerlt and Gassman have applied Marcus formalism to such proton transfer reactions to argue that transition states for concerted general acid-general base catalysed enolization at enzyme active sites occur late on the reaction coordinate (J. Am. Chem. Soc. 115 (1993) 11552). We postulate that as a...

Pentavalent Organo-Vanadates as Transition State Analogues for Phosphoryl Transfer Reactions.

Pentavalent organo-vanadates have been put forth as transition state analogues for a variety of phosphoryl transfer reactions. In particular, uridine 2',3'-cyclic vanadate (U>v) has been proposed to resemble the transition state during catalysis by ribonuclease A (RNase A). Here, this hypothesis is tested. Lys41 of RNase A is known to donate a hydrogen bond to a nonbridging phosphoryl oxygen in...

Elementary Steps in Enzyme Catalysis and Regulation

A general approach to elucidating the molecular basis of enzyme catalysis and regulation is the time resolution of the complex reaction mechanisms involved into their elementary steps. Both enzyme catalysis and enzyme regulation are initiated by the binding of small molecules to the protein. This association reaction is generally quite rapid and i often followed by a conformational change of th...
